Validation of Diagnostic Markers for Streak Virus Disease Resistance in Maize

Maize streak virus (MSV) is responsible for disease of maize and poses a serious threat to production in sub-Saharan Africa. Polygenic resistance MSV has become an essential requirement modern cultivars mitigate yield losses. Many single nucleotide polymorphism (SNP) markers linked putative loci have been identified use forward breeding. This study aimed validate, using the high-throughput kompetitive allele specific PCR (KASP) assay, diagnostic ability three SNP Msv1 trait 151 early generations inbred lines with diverse genetic backgrounds, together nine MSV-resistant elite susceptible check (cv. Pool-16). The phenotypic responses were determined by inoculation viruliferous leafhoppers (Cicadulina triangular) under screenhouse conditions. Based on established rating system, categorized into resistant, moderately susceptible, highly susceptible. SNPs associated detected 133 lines, which as resistant (54), (76), (1). 18 generation without these classified (10), (5), (3). confirms strong association their usefulness breeding while emphasizing need identify additional screen any ambiguity.
